In Ca2+-free recoverin, the myristoyl group (C14:0) is sequestered in a hydrophobic pocket formed by EF1, EF2 and EF3. Significant conformational change occurs in these EF-hands upon binding of Ca2+ at EF2 and EF3, leading to the ejection of the myristoyl group, thus enabling recoverin to bind to membranes. EF4 undergoes little conformational change, as its interaction with the C-terminal K-helix of recoverin locks it in a permanent open conformation. |
